I believe socialtoo (http://www.socialtoo.com) has an unfollow tool, and various auto-follow capabilities as well. Robert Scoble used it to unfollow his massive list of tweeps a few months ago (including me, sadly).

'Your Twitter Karma' no longer allows the bulk unfollow option, as it violates Twitter policy or some such. I just found this out today when trying to delete my non-followers.

This will eventually become the norm unfortunately...
